【摘要】目的探討干擾素(IFN)對人結(jié)腸癌細(xì)胞人類白細(xì)胞抗原DR(HLA-DR)表達(dá)的影響。
方法采用免疫組化SP法和ELISA法分別檢測不同劑量的α-IFN和γ-IFN刺激前、后HLA-DR在4株人結(jié)腸癌細(xì)胞HCT-8、Lovo、SW-480和Ls-174-T中的表達(dá)情況。
結(jié)果α-IFN和γ-IFN刺激之前,4株細(xì)胞中除Lovo細(xì)胞不表達(dá)HLA-DR,其他3株細(xì)胞均可不同程度表達(dá)HLA-DR; 刺激之后,4株細(xì)胞系的HLA-DR表達(dá)均有所增強(qiáng),并呈現(xiàn)出IFN依賴性及γ-IFN的劑量依賴性,γ-IFN的誘導(dǎo)作用強(qiáng)于α-IFN。
結(jié)論IFN可以上調(diào)人結(jié)腸癌細(xì)胞HLA-DR的表達(dá),IFN用于結(jié)腸癌的臨床治療具有一定的應(yīng)用前景。
