目的觀察脂質(zhì)體介導(dǎo)增殖細(xì)胞核抗原（PCNA）對(duì)肝癌細(xì)胞增殖的影響。 方法根據(jù)PCNA cDNA序列設(shè)計(jì)合成與PCNA mRNA AUG起始密碼子后的18位堿基序列互補(bǔ)的PCNA反義寡核苷酸，脂質(zhì)體介導(dǎo)轉(zhuǎn)染人肝癌細(xì)胞株，通過細(xì)胞生長曲線測定，MTT比色法檢測PCNA反義寡核苷酸對(duì)人肝癌細(xì)胞的生長抑制率，比較陽離子脂質(zhì)體介導(dǎo)轉(zhuǎn)染法與直接轉(zhuǎn)染法的區(qū)別。 結(jié)果PCNA反義寡核苷酸作用后的肝癌細(xì)胞，生長明顯受到抑制。應(yīng)用陽離子脂質(zhì)體介導(dǎo)轉(zhuǎn)染能顯著提高PCNA反義寡核苷酸的抑增殖效應(yīng)。結(jié)論利用陽離子脂質(zhì)體介導(dǎo)轉(zhuǎn)染PCNA反義寡核苷酸能顯著提高對(duì)人肝癌細(xì)胞增殖的抑制效應(yīng)。
