為觀察促肝細胞生長素(pHGF)對肝硬變大鼠部分肝切除后肝再生的影響,應(yīng)用IBASⅡ全自動圖像分析儀檢測肝硬變大鼠術(shù)后肝細胞DNA倍體率及肝臟組化染色后各組酶灰度值(OPTDM)的變化,并通過檢測血清谷丙轉(zhuǎn)氨酶(SGPT)及吲哚青綠15分鐘滯留率(ICG15)以了解殘肝功能。結(jié)果:實驗組(A組)3~4倍體肝細胞明顯減少,2倍體及多倍體肝細胞明顯增多;術(shù)后第1天各組酶灰度值無顯著差異,第2、5天琥珀酸脫氫酶(SDH)和乳酸脫氫酶(LDH)明顯增高,而堿性磷酸酶(AkP)和酸性磷酸酶(AcP)則明顯減少;A組術(shù)后第5天SGPT和ICG15明顯降低。本實驗結(jié)果提示:pHGF不僅能促使肝硬變大鼠肝切除后殘肝的再生,而且能夠促進再生肝細胞功能成熟及殘肝功能的恢復(fù)。
