目的 觀察阿魏酸鈉對肺纖維化大鼠肺組織轉(zhuǎn)化生長因子β1(TGF-β1)信號通路分子mRNA表達的影響,探討其防治肺纖維化的作用及機制。方法 氣管內(nèi)滴入博來霉素(5 mg/kg)建立大鼠肺纖維化模型。將30只SD大鼠隨機分為三組(每組10只):對照組、肺纖維化模型組和阿魏酸鈉組。HE染色檢測肺組織病理改變,膠原纖維染色檢測膠原纖維表達,原位雜交技術(shù)檢測肺組織TGF-βRⅡ及Smad4 mRNA表達,實時熒光定量RT-PCR技術(shù)檢測TGF-β1 mRNA表達。結(jié)果 膠原纖維染色顯示模型組大鼠肺內(nèi)膠原纖維表達顯著高于對照組和阿魏酸鈉組(P lt;0.001)。大鼠肺內(nèi)TGF-β1、TGF-βRⅡ和Smad4的mRNA表達在模型組顯著高于對照組(P lt;0.01),在阿魏酸鈉組顯著低于模型組(P lt;0.05)。結(jié)論 阿魏酸鈉能有效地減輕肺纖維化大鼠肺組織的纖維化程度,其作用機制主要是通過抑制TGF-β1、TGF-βRⅡ和Smad4的mRNA表達上調(diào),從而干擾TGF-β1/Smad4信號通路對靶基因的激活來實現(xiàn)的。
