目的  探討不可分型流感嗜血桿菌(NTHi)作用于呼吸道上皮細(xì)胞株(A549),表達(dá)前炎癥細(xì)胞因子的變化及其干預(yù)措施。方法  用NTHi、NTHi+紅霉素(10 mg/L)、NTHi+慶大霉素(100 mg/L)、NTHi+地塞米松(100 μmol/L)分別感染A549細(xì)胞,用核因子κB(NF-κB)抑制劑二硫氨基甲酸吡啶(PDTC,200 μmol/L)預(yù)處理NTHi感染的A549細(xì)胞,24 h后觀察細(xì)胞的形態(tài)學(xué)改變,并檢測(cè)上清液中白細(xì)胞介素8(IL-8)、腫瘤壞死因子α(TNF-α)的水平和細(xì)胞間黏附分子1(ICAM-1)的表達(dá)。結(jié)果  NTHi作用A549細(xì)胞24 h后,引起部分上皮細(xì)胞變形、死亡。NTHi能顯著誘導(dǎo)A549細(xì)胞株分泌IL-8、表達(dá)ICAM-1,紅霉素能夠抑制其誘導(dǎo)作用;慶大霉素具有殺滅NTHi作用,但不能抑制IL-8分泌和ICAM-1表達(dá);地塞米松能抑制A549分泌IL-8,降低ICAM-1的表達(dá),但不能殺滅NTHi。PDTC(200 μmol/L)能阻斷A549細(xì)胞在NTHi作用后IL-8的高表達(dá)。TNF-α在各組培養(yǎng)上清中均不能檢測(cè)到。結(jié)論  NTHi顯著誘導(dǎo)人肺上皮細(xì)胞系表達(dá)前炎癥因子,該作用可能通過(guò)NF-κB通路。紅霉素在殺滅NTHi的同時(shí)還具有抗炎作用。
