目的 根據(jù)基因庫中的人肝素酶(HPSE)基因序列,構(gòu)建針對肝素酶基因的小干擾RNA(siRNA)及其表達載體,轉(zhuǎn)染MDA-MB-231細胞后觀察其對肝素酶基因的干擾作用。
方法 設計肝素酶靶向的發(fā)夾狀siRNA,合成2條互補的寡核苷酸鏈共4對,退火后連接入pGPU6/GFP/Neo載體,轉(zhuǎn)化后進行序列鑒定,轉(zhuǎn)染MDA-MB-231細胞,熒光照片,RT-PCR (real-time PCR)檢測肝素酶mRNA的表達情況。 結(jié)果 設計出4條針對肝素酶的siRNA,將退火后的雙鏈寡核苷酸片段克隆到pGPU6/GFP/Neo載體,經(jīng)過陽性菌落酶切鑒定與測序,結(jié)果正確。再將siRNA載體轉(zhuǎn)染到MDA-MB-231細胞株,RT-PCR檢測肝素酶mRNA表達明顯降低(P lt;0.05)。
結(jié)論 成功構(gòu)建了針對肝素酶基因的siRNA載體,轉(zhuǎn)染到MDA-MB-231細胞株后細胞肝素酶基因的表達明顯降低,從而為腫瘤治療提供了一種新的方法。
