【摘要】目的探討三七總皂苷(PNS)預處理對大鼠供肝缺血-再灌注損傷的保護作用及其對供肝細胞凋亡和Bcl-2及Caspase-3 mRNA表達的影響。
方法雄性SD大鼠分別用作供、受體,采用Kamada’s袖套法建立原位肝移植模型,根據(jù)供肝切取前1 h是否靜脈注射PNS(50 mg/kg)將大鼠隨機分為PNS預處理組(PNS組)和生理鹽水對照組(NS組); 另設假手術作對照組(SO組)。分別于供肝再灌注后 2 h、6 h及24 h處死各組動物,檢測血清ALT及AST,HE切片作病理組織學檢查,TUNEL法檢測肝細胞凋亡,RT-PCR法檢測Bcl-2及Caspase-3 mRNA的表達。
結果供肝再灌注后2 h、6 h及24 h各時點,PNS組大鼠血清ALT和AST水平及肝細胞凋亡指數(shù)(AI)均明顯低于NS組(P<0.05,P<0.01); 6 h及24 h,PNS組大鼠肝組織Bcl-2 mRNA的表達水平明顯高于NS組(P<0.05); 2 h及6 h,PNS組大鼠肝組織Caspase-3 mRNA的表達水平明顯低于NS組(P<0.05)。
結論PNS預處理大鼠供肝,可以有效地減輕移植肝的缺血/再灌注損傷和細胞凋亡,影響細胞凋亡調控基因Bcl-2和 Caspase-3的表達。這可能為PNS抗細胞凋亡的機理之一。
